Genetic isolation is the absence of genetic exchange between populations or species as a result of geographic separation or of mechanisms that prevent reproduction.
The distance is the main factor for genetic isolation. Due to the enhanced technology and transportation methods, genetic isolation among humans are diminishing.
But for other species it will matter since they cannot reach another region easily. For example a deer from the USA will have problems getting to Europe because of the sea. (Back in the days it was assumed that the continents stayed together and then moved apart)
Human beings and thier environmental impacts could play a role too since environmental pollution is a main cause for endangering animal and plant species. Therefore some species can only survive in certain parts of the world and gets isolated into these regions due to human activities.
The animals gets used to the environment and weather in those regions and cannot survive in a different environment and thereby isolates themselves to a specific region away from its ancestors.
Those are the only ones I can think of so far.
