How is isolation of gene of interest carried out?

please suggest me how to carry out isolation of gene of interest using restriction endonucleases ......also send me the site from which it is available???How is isolation of gene of interest carried out?
%26gt;isolation involves following steps...





1%26gt;breaking or opening of the cells to expose nucleic acid.


2%26gt;separation of nucleic acid from other cellular component.


3%26gt;recovery of nucleic acid in pure form.


Now to obtain the fregment of DNA containing desired gene





%26gt;specific restriction enzymes are used to cut the DNA at a specific place.





%26gt;the cut DNA fregment may have mostly sticky ends or blunt ends, sticky ends these are particularly useful for rDNA experiment.





%26gt;these cut DNA fregment are joined with the vectors DNA with the help of DNA ligase enzyme.
